Thermo Scientific DyNAmo SNP Genotyping Master Mix delivers fast, high quality SNP genotyping to customers using end-point fluorescence detection on Real-Time PCR instruments. Reliable and reproducible detection of SNP alleles is achieved via a patent pending master mix formulation that offers enhanced discrimination, even when targeting the most challenging amplicons. DyNAmo SNP Genotyping Master Mix has been optimized to offer superior performance when using TaqMan SNP Genotyping Assays or custom designed assays.

DyNAmo SNP Genotyping Master Mix delivers superior performance also with challenging amplicons

The performance of DyNAmo SNP Genotyping Master Mix was compared against three other industry leading master mixes specifically developed for genotyping protocols. All Master Mixes tested delivered an acceptable call rate for an assay targeting a LAC2 SNP (rs4988235) but for a more challenging amplicon (NOTCH2 SNP; rs2453042), two of the four mixes failed to successfully call the correct genotypes.

Accurate genotyping results over a range of template concentrations

Allelic discrimination plot of 12 human genomic DNA samples, generated using a custom designed hydrolysis probe assay targeting a SNP in NOTCH2 (rs2453042), and DyNAmo SNP Genotyping Master Mix. DyNAmo SNP Genotyping Master Mix has been demonstrated to provide accurate genotyping results within a range of 1-10 ng of source DNA, and in this particular data set, accuracy was maintained with an input level of 0.5 ng of gDNA.

Achieve improved call rates compared to non-optimized master mix for probe chemistry

The performance of DyNAmo SNP Genotyping Master Mix versus a non-optimized master mix for probe chemistry was assessed. Allelic discrimination plots displayed were generated from 21 human genomic DNA samples (5 ng per sample), using custom designed hydrolysis probe assays targeting a SNP in NOTCH2 and TXB18 (rs2453042 and rs215938, respectively). Although comparable call rates were observed when comparing TXB18 data generated from the two different master mixes detailed, when using the non-optimized master mix, analysis of the NOTCH2 SNP resulted in unusable data. Optimized DyNAmo SNP Genotyping Master Mix offers more consistent results when compared to master mixes not optimized specifically for genotyping applications.

Shipping and storage

DyNAmo SNP Genotyping Master Mix is shipped on gel ice. Upon arrival, store all the components at -20°C. When using the 2x master mix, the leftover thawed mix can be refrozen and stored at -20°C without affecting the performance of the product.
